IMO, the TOUD will focus at least in part on one of the major conflicts that I see brewing around Strahd. The latest reincaration of Tatyana (aka Tara Kolyana) has escaped his grasp, dodging her own prophetic doom and living far longer than she was suppose to. In the process she has taken on the mantle of an Anchorite, and--unlike any Tatyana incarnation before her--has begun recalling events from her past lives without the aid of Strahd's hypnosis.
In IStWAA, Azalin comments that Strahd's greatest weakness may be his attraction to Tatyana, and spectulates as to how this could be turned against him. I can see Azalin trying to manipulate Tara against Strahd and finding out she's more than he can handle.
Bring in the GC and Van Richten into this mess and you'd have a free-for-all of epic proportions.
